POSTECH·교토대, 신약 개발 정확도 높일 ‘오가노이드칩’ 기술 동향 총정리

POSTECH 기계공학과 윤정빈 연구조교수와 기계공학과·IT융합공학과·융합대학원·생명과학과 장진아 교수 연구팀은 일본 Kyoto University 류지 요코가와 교수 연구팀과 함께 신장 및 방광 오가노이드칩 기술의 최신 연구 동향을 분석한 리뷰 논문을 국제 학술지 Advanced Drug Delivery Reviews에 게재했다고 밝혔다.

 

신장과 방광은 체내에서 약물의 흡수와 배설, 독성 반응 등에 중요한 역할을 담당하는 기관이다. 그러나 기존의 세포배양 실험이나 동물실험만으로는 실제 인체에서 나타나는 반응을 정확하게 예측하는 데 한계가 있어 신약 개발 과정에서 예상치 못한 부작용이 발생하는 경우가 적지 않았다.

 

특히 약물로 인한 신장 손상은 임상시험 실패의 대표적인 원인으로 꼽히며, 보다 정교한 전임상 평가 시스템의 필요성이 지속적으로 제기돼 왔다.

 

연구팀은 이번 논문에서 신장과 방광을 모사하는 미세생리시스템(MPS)의 발전 과정을 체계적으로 분석했다. 초기 2차원 세포배양 기술부터 오가노이드 기반 장기칩, 혈관화 기술, 실시간 모니터링 센서, 3차원 바이오프린팅, 인공지능(AI) 기반 자동화 시스템까지 관련 기술의 진화 과정을 폭넓게 정리했다.

 

오가노이드칩은 인간 줄기세포를 활용해 만든 미니 장기(오가노이드)를 미세유체칩과 결합한 기술로, 실제 인체 장기의 구조와 기능을 실험실 환경에서 보다 정밀하게 재현할 수 있다는 점에서 주목받고 있다.

 

연구팀은 특히 신장과 방광을 개별적으로 분석하는 수준을 넘어 두 장기의 상호 연관성을 고려한 통합 평가 시스템의 중요성을 강조했다. 약물의 배설 경로와 체내 노출 정도를 보다 현실적으로 예측할 수 있어 신장 독성 평가뿐 아니라 방광 내 약물 전달 효과 분석에도 활용 가능성이 크다는 설명이다.

 

또한 오가노이드칩에서 얻은 데이터를 생리학 기반 약동학 모델(PBPK)이나 디지털 트윈 기술과 연계할 경우 신약 후보물질의 효과와 부작용을 더욱 정밀하게 예측할 수 있을 것으로 전망했다. 이는 신약 개발 과정의 시행착오를 줄이고 전임상 시험의 신뢰성을 높이는 데 기여할 수 있다는 평가다.

 

윤정빈 연구조교수는 “이번 연구는 신장과 방광 장기칩 기술이 현재 어느 수준까지 발전했는지와 앞으로 해결해야 할 과제를 종합적으로 정리한 데 의미가 있다”며 “국제 공동연구를 통해 사람 중심의 차세대 전임상 평가 기술 발전 방향을 제시했다”고 말했다.

 

한편 이번 연구는 과학기술정보통신부와 교육부의 지원을 받아 한국연구재단 세종과학펠로우십 사업 및 글로벌 헬스케어 의공학 연구소(국가연구소사업)를 통해 수행됐다.

POSTECH and Kyoto University Summarize Trends in ‘Organoid Chip’ Technology to Enhance Accuracy in New Drug Development

 

The results of an international joint research project summarizing the development direction of ‘Organoid-on-a-Chip,’ a technology garnering attention as a next-generation preclinical evaluation tool to resolve the recurring clinical trial failures in the new drug development process, have been announced.

 

A research team led by Research Assistant Professor Jungbin Yoon of the Department of Mechanical Engineering at POSTECH and Professor Jinah Jang of the Department of Mechanical Engineering, Department of IT Convergence Engineering, Graduate School of Convergence, and Department of Life Sciences announced that they have published a review paper analyzing the latest research trends in kidney and bladder organoid chip technology in the international academic journal Advanced Drug Delivery Reviews, in collaboration with a research team led by Professor Ryuji Yokogawa of Kyoto University in Japan.

 

The kidneys and bladder are organs that play crucial roles in the body regarding drug absorption, excretion, and toxic reactions. However, existing cell culture or animal experiments have limitations in accurately predicting reactions that occur in the actual human body, leading to a significant number of unexpected side effects during the new drug development process. In particular, drug-induced kidney damage is cited as a leading cause of clinical trial failure, and the need for more sophisticated preclinical evaluation systems has been continuously raised.

 

In this paper, the research team systematically analyzed the development of microphysiological systems (MPS) that mimic the kidneys and bladder. They broadly summarized the evolution of related technologies, ranging from early 2D cell culture techniques to organoid-based organ chips, angiogenesis technology, real-time monitoring sensors, 3D bioprinting, and AI-based automation systems.

 

Organoid chips are a technology that combines mini-organs (organoids) created using human stem cells with microfluidic chips; they are attracting attention for their ability to more precisely reproduce the structure and function of actual human organs in a laboratory setting.

 

The research team particularly emphasized the importance of an integrated evaluation system that considers the interrelationship between the two organs, going beyond the level of analyzing the kidneys and bladder individually. They explained that because it allows for more realistic prediction of drug excretion pathways and the degree of exposure within the body, it has great potential for use not only in evaluating kidney toxicity but also in analyzing drug delivery effects within the bladder.

 

Furthermore, it is projected that integrating data obtained from organoid chips with physiology-based pharmacokinetic (PBPK) models or digital twin technology will enable more precise prediction of the efficacy and side effects of new drug candidates. This is evaluated as contributing to reducing trial and error in the drug development process and enhancing the reliability of preclinical trials.

 

Research Assistant Professor Yoon Jung-bin stated, "This study is significant in that it comprehensively summarizes the current level of advancement in kidney and bladder organ-chip technology and the challenges that need to be addressed in the future," adding, "Through international collaborative research, we have presented a direction for the development of human-centered next-generation preclinical evaluation technologies."

 

Meanwhile, this research was conducted with support from the Ministry of Science and ICT and the Ministry of Education through the National Research Foundation of Korea's Sejong Science Fellowship Program and the Global Healthcare Biomedical Engineering Institute (National Research Institute Project).
